The Czech Advanced Light and Electron Microscopy Multi Modal Multi Sited Node Candidate in Prague offers a wide range of state-of-the-art light and electron microscopy equipment and techniques. The Node Candidate is also active in organizing training and courses focused on theoretical and practical aspects of basic and advanced microscopy techniques. Light microscopy instrumentation includes microscopy systems that range from routine fluorescence microscopes, confocal microscopes up to high-end super-resolution microscopy systems. In the area of electron microscopy, the Node Candidate provides expertise and cutting edge equipment for a broad range of biological sample preparation and ultrastructural imaging techniques, including cryo techniques, electron tomography, and electron energy filtering.
Specialties and expertise of the Node Candidate
The Advanced Light Microscopy (ALM) core facility at IMG CAS is focused on cutting-edge microscopy techniques particularly on time-gated STED microscopy. It provides strong support in image analysis and processing, more specifically in image reconstruction and deconvolution, image registration, volume reconstruction, image segmentation, tracking, colocalization analysis, analysis of photokinetic experiments, 3D visualization and mathematical modeling and simulations of biological processes.
The Electron Microscopy (EM) core facility at IMG CAS has a long expertise in the development and optimization of sample preparation techniques for electron microscopy, e.g. development of a new system for simultaneous immunolabeling of multiple molecular targets, a special technique for combination of 3D pre-embedding immunolabeling and 2D on-section immunolabeling, a statistical tool for evaluation of clustering and colocalization of immunolabeling in EM.
The Imaging core facility at BIOCEV focuses on fluorescence super-resolution imaging, functional imaging, two-photon imaging and scanning electron microscopy. The strength in image analysis is especially in fluorescence lifetime imaging data analysis and in spatio-temporal correlation techniques, for which unique data analysis modules were developed.
Image analysis and stereology is an area of expertise of the IPHYS BioImaging Facility. It provides development of new methods and SW modules (in ImageJ, Amira or Ellipse SW environment) in the field of image processing and analysis, stereology and spatial statistics.

Leica TCS SP8 STED 3X microscope is state-of-the-art 3D time gated STED confocal imaging platform offering resolution up to 50nm in lateral and 130nm in axial direction.
In EM, special features of the instruments are availability of the instruments for sample preparation by cryofixation and the microscope equipment allowing for electron tomography, cryomicroscopy and electron energy filtering.
